Chloroquine CQ and hydroxychloroquine HCQ, two antimalarial drugs, are suggested to have potential anticancer properties. in the present study, we investigated the effects of CQ and HCQ on cell growth of bladder cancer with emphasis on autophagy inhibition and apoptosis induction in vitro. Chloroquine also obviously weakened the repair of radiation-induced DNA damage. A combination of radiation and chloroquine enhanced the apoptosis rate of EJ and T24 cells and down-regulated the expression of Bcl-2 while up-regulating the expression of caspase-3. Chloroquine CQ is a lysosomotropic agent with an extensive range of biological effects 1. Historically known for its anti-malarial activity, chloroquine is a widely used biological research tool for studying autophagy inhibition. Research studies demonstrate that chloroquine accumulates in acidic lysosomes and increases the lysosomal pH.

For all uses of chloroquine: WARNING/CAUTION: Even though it may be rare, some people may have very bad and sometimes deadly side effects when taking a drug. Do not start, stop, or change the dose of any drug without checking with your doctor.

  7. Introduction. Chloroquine is an antimalarial drug that has been used in humans for many years 1. In recent years, Chloroquine has been shown to inhibit autophagy and induce apoptosis in malignant cells and thus has been tested in various experimental model systems 2 and in human clinical trials 3, 4.

The combination of chloroquine with zinc enhanced chloroquine’s cytotoxicity and induced apoptosis in A2780 cells. Thus chloroquine is a zinc ionophore, a property that may contribute to chloroquine’s anticancer activity. Chloroquine inhibits the intracellular multiplication of Legionella pneumophila by limiting the availability of iron. The chloroquine-induced increase in the expression levels of p53 target genes was also observed in other cell lines expressing wtp53 such as HCT116 and G168 Fig. 3B, right panel and data not shown. In contrast, chloroquine treatment did not induce p53 target genes in glioma lines lacking functional p53 Fig. 3C. In Myc-driven lymphomas in which tumor regression was initiated with either p53 activation or alkylating chemotherapy, either chloroquine treatment or knockdown of the essential autophagy gene ATG5 resulted in a similar enhancement of therapy-induced apoptosis. Chloroquine treatment of cells deficient in autophagy did not further enhance cell death, suggesting that, at low micromolar concentrations, the anticancer effects of chloroquine were due to its action as an autophagy inhibitor.
