Side effects are generally mild and transitory, although visual disturbance, muscle weakness, and acute psychotic behavior have been encountered.

    The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

    If possible, suppressive therapy should start 2 weeks prior to exposure; if unable to start 2 weeks before exposure, an initial loading dose of 1 g chloroquine phosphate 600 mg base may be taken orally in 2 divided doses, 6 hours apart. -Suppressive therapy should continue for 8 weeks after leaving the endemic area. The therapeutic dose of chloroquine phosphate is 500 mg once a week for malaria prophylaxis or 2.5 g over 2 days for treatment of malaria. Deaths have been reported in children after ingesting one or two tablets—doses as low as 300 mg; the lethal dose of chloroquine for an adult is estimated at 30–50 mg/kg. Chloroquine is an aminoquinoline used for the prevention and therapy of malaria. It is also effective in extraintestinal amebiasis and as an antiinflammatory agent for therapy of rheumatoid arthritis and lupus erythematosus. Chloroquine is not associated with serum enzyme elevations and is an extremely rare cause of clinically apparent acute liver injury.
