    Chloroquine glutathione synthase

Chloroquine has been extensively used in mass drug administrations, which may have contributed to the emergence and spread of resistance. It is recommended to check if chloroquine is still effective in the region prior to using it.

Oct 01, 2018 The dosage of Chloroquine phosphate is often expressed in terms of equivalent Chloroquine base. Each 500 mg tablet of Chloroquine phosphate contains the equivalent of 300 mg Chloroquine base. In infants and children the dosage is preferably calculated by body weight. Glutathione is called an antioxidant because of its role in protecting cells from the damaging effects of these unstable molecules. Mutations in the GSS gene prevent cells from making adequate levels of glutathione, leading to the signs and symptoms of glutathione synthetase deficiency. The genes encoding enzymes involved in glutathione GSH metabolism may modulate responses to antimalarial drugs, but the role of most of them in antimalarial drug resistance has scarcely been investigated.

The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

  5. The systematic name of this enzyme class is gamma-L-glutamyl-L-cysteineglycine ligase ADP-forming. Other names in common use include glutathione synthetase, and GSH synthetase. This enzyme participates in glutamate metabolism and glutathione metabolism. At least one compound, Phosphinate is known to inhibit this enzyme.

Glutathione is held in a reduced state by glutathione reductase and thioredoxin. Conditions of high oxidative stress lead to depletion of glutathione, which, in turn, increases the biosyn-thetic activity of γ-glutamylcysteine synthase to restore glutathione to normal levels. Keeping that statement in mind I will address foods that can aid in glutathione synthesis. The first step in producing glutathione is catalyzed by gamma-glutamylcystein synthase. This enzyme binds cysteine to the gamma carbon of glutamate which forms gamma-glutamylcystein. Mutants lacking glutathione synthetase, which is the second enzyme of glutathione synthesis and is encoded by the GSH2 gene, have no GSH and are not viable. However, gsh2 mutants can be fully complemented by expression of GSH2 in the cytosol Pasternak et al. 2008, showing that expression in the cytoplasm is sufficient to restore the wild‐type growth phenotype.
