Although the mechanism of action is not fully understood, chloroquine is shown to inhibit the parasitic enzyme heme polymerase. Chloroquine phosphate, USP is an antimalarial and amebicidal drug. Each tablet, for oral administration, contains 250 mg of chloroquine phosphate, USP equivalent to 150 mg base. It is not effective against exoerythrocytic forms of the parasite.

The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.
