Chloroquine is a 4-aminoquinoline with antimalarial, anti-inflammatory, and potential chemosensitization and radiosensitization activities. Although the mechanism is not well understood, chloroquine is shown to inhibit the parasitic enzyme heme polymerase that converts the toxic heme into non-toxic hemazoin, thereby resulting in the accumulation of toxic heme within the parasite. That would be the synthesis of chloroquine from 4,7-dichloroquinoline and 4-diethylamino-1-methylbutylamine, where both starting materials are available through well-known procedures. This thesis describes research targeted at the modification of lead reversed chloroquine molecules to discover new and effective moieties, as well as to improve pharmacokinetic-related properties. An especial emphasis of this project is the addition of a sulfonamide functional group to a reversed chloroquine.

Synthesis of benzotriazole Medicinal Chemistry BACKGROUND Principle Benzotriazole can be prepared by treating o-phenylenediamine with nitrous acid liberated during the reaction between sodium nitrite. Design and Synthesis of Autophagy Inhibitors Based on the Structure of the Antimalarial Drug Chloroquine Amaravadi Laboratory, University of Pennsylvania School of Medicine The development of autophagy autophagocytosis inhibitors represents an important approach to the potentiation of the effects of cancer chemotherapeutic agents. During the literature search, it was observed that there are only two approaches for the synthesis of chiral chloroquine using glutamic acid and pyroglutamic acid as starting materials.7, 8 There are limitations associated with these methods viz. the reaction involves the conversion of carboxylic acid group to methyl which is a low yielding step, secondly, only methyl group could be introduced at the chiral center. In order to optimize substituent group at the chiral carbon, it is important.

Chloroquine and primaquine combining old drugs as a new. Primaquine is active as a chloroquine-resistance reverser at concentrations that could be clinically relevant. As primaquine and chloroquine are already used together in the treatment of P. vivax, this is potentially a cheap way of reviving and extending the life of chloroquine.

PLaquenil and drinking alcohol - Scleroderma - Inspire I have taken plaquenil since 1989 and have had no problem with the drug. I only get my eyes examined once a year now, as the Dr. felt I am tolerating the drug without complication. I am wondering though, if anyone also takes minocin or minocycline along with the plaquenil.
